There are many types of aero wheels out there, but which is faster?

*Blade carbon spoke (HED, Spinergy, etc.)
*Deep sectioned wheels. And what rim depth get the most bang for the dish?

How fast do you have to go to get a significant increase in speed?

The Hed# has shown to have the least drag at 30 mph in the wind tunnel (aside from a flat disk). However, realize that a disk, and even the Hed3, is going to more effected by crosswinds. This is why disks aren't allowed to be used as front wheels in tris. The Hed3 is allowed, and I use them, but in very windy races, or in mass start races (tris aren't mass start), I don't use the Hed3 front. You'll notice a difference in basically anything over 16 mph. Do some searching around the Slowtwitch site for more wheel info.
