Thanks to all who answered my earlier questions. Next question:

In reading the replies and checking out some maps, I want to do a road ride along Skyline Drive at least once (that may be all I can get away with). If I were going to drive to one spot from which to climb up to Skyline and I wanted to pick the most epic climb I could, where would I park and which road would I take to get up to Skyline? Some of them appear to have quite a few switchbacks which is usually a good sign.

There are no real epic climbs, or many switchbacks, but the riding is great. You can make a nice 60 miles roundtrip to the next highway intersection with Skyline. Next few weeks are probably not too safe due to fall color tourists.
38/26 makes an easy spin, and the climbs are never ending and the mileage markers every mile drive you nuts.

try Thorn Gap (US 211) or (US 33), they're nice climbs. If you wanna get good climb park at New Market and ride up to the top on 211. ROad is busy but there's enough schoulder room.
